Will it work if the fileformat is in ".xls"?

Yes, the property Workbook.IsPasswordProtected(fileName) could work for .xls fileformat. Please download Spire.XLS Pack(Hotfix) Version:9.10.4 and have a try.
If you still have the issue, please provide your protected file along with the password for testing.
